Janus kinase 2 gene (JAK2) mutations were found in several myeloproliferative neoplasms (MPNs), most frequently in polycythemia vera (about 100%), and nearly in one-half of essential thrombocythemia and primary myelofibrosis. JAK2 mutations were reported to cause constitutive activation of an intracellular tyrosine kinase, in many hematopoietic cells. This test has diagnostic value as it distinguishes non-CML MPNs. JAK2- mutations were also reported to have association with Budd-Chiari syndrome (BDCHS).
